SCOPE

Identifying disease-causing genes represents a major focus in current medical genetics research, and next-generation sequencing (NGS) technology is profoundly changing the way of diagnostics and therapeutics, especially in oncology. Coping with the underlying informational complexity in precision medicine requires multidisciplinary efforts that generate synergies through cross-disciplinary research at the nexus of medicine, biology, and informatics.

 

With this Summer School, we address precisely this challenge and cover the entire process from clinical phenotyping, to NGS data analysis, to clinical and genetic follow-up, to finally guide development of therapies and (personalized) clinical applications.

 

In particular, we will focus on hereditary cancer predisposition syndromes, sporadic cancer disease and novel therapeutic approaches with a special focus on resources available in Tübingen. The concept of Precision Medicine and interdisciplinary approaches within the Center For Personalized Medicine will be part of the presentation as well as the integration of complex omics technologies.

 

This Summer School is tailored for oncologists with an interest in science, scientists working in the diagnostic field as well as post-docs and PhD students working in the fields of biology, neuroscience, biochemistry, experimental medicine, and bioinformatics.

 

Participants of the 3-day-event held in English will receive 3 ECTS credit points according to the European Credit Transfer System for their attendance of the Summer School and presentation of a poster. For physicians, the event is acknowledged with 25 CME (Category A) points.

KEYNOTE LECTURE

Molecular Insights via the Analysis of Large-scale (Cancer) Genomics Data

Pan-cancer studies have transformed our understanding of recurrent somatic mutations that contribute to cancer pathogenesis; however, there has yet to be a full investigation of the multiple mechanisms in which genes can be somatically altered.

 

Here, we present a comprehensive catalogue of cancer-associated gene alterations through the extensive characterization of tumor transcriptomes from ≈1,200 donors of the PanCancer Analysis of Whole Genomes (PCAWG) project with matched whole-genome sequence data. We identified gene alterations through major gene expression changes, alternative splicing, alternative transcription starts, allele-specific expression, RNA-edited sites, and gene fusions. Our analysis shows that key cancer genes are frequently affected on the RNA level even when there is no direct evidence of a functional DNA alteration (e.g., coding SNV or CNA). In many cases we can statistically link cis somatic mutations to identified RNA alterations. In a novel recurrence analysis, we also identify novel genes that are recurrently altered at DNA or RNA level and that may play important roles in tumorigenesis. We also report on the analysis of splicing aberrations in more than 8,000 tumors across 30 cancer types (TCGA PanCanAtlas). We find distinct differences between tumor and normal splicing and that 10-20% of the samples exhibit a very high burden of splicing aberrations. In those samples we identify many new splice junctions in coding regions that may lead to many new translated peptides that likely have major implications for immunotherapy.

 

The talk will also cover recent work on the joint analysis of clinical text notes and somatic mutations and our data sharing efforts in the context of the Global Alliance of Genomics and Health (GA4GH) that has lead to the largest public repository of BRCA1/2 variants (brcaexchange.org, currently 18,960 unique variants).

  • By train to Tübingen main station. Tübingen's main station is centrally located on the southern side of the town centre. It is well connected to the ICE train station Stuttgart. For more information please check: www.bahn.de
  • By plane to Stuttgart airport (STR). The airport sprinter shuttle 828 connects Stuttgart airport with Tübingen, total duration: ~1h,
    costs: 7 EUR one way, timetable available here.
  • By car. Tübingen is located on the intersection of the B27 and B28 and has fast connections to the A8 (Munich-Stuttgart) and the A81 (Singen-Heilbronn). There is paid on-site parking available. We do however recommend to arrive by public transport and leave your car at the hotel.
